I think that there were many factors that led to Hitler's consolidation of power, including a seeming and unfathomable acquiescence by the general population. I was not previously aware, or had forgotten, about the Night of the Long Knives, and did a quick look/see on the Internet before responding. Since it (the planned removal exercise to remove opposition even within the Nazi Party itself) happened relatively late (June 1934), it is hard to accept it as "the" defining moment. We can only hope no future despot will assume dictatorial power with such tragic consequences.
